ABSTRACT

Resumen Introducción. Haematococcus pluvialis es una microalga que produce astaxantina, un beta-caroteno y antioxidante muy usado en la industria. Para obtener una mayor producción de astaxantina se planteó como objetivo utilizar diferentes factores de estrés, en un biorreactor a escala de laboratorio de 5 litros. Metodología. Se cultivó la microalga en el medio RM, pH 6,8, temperatura 20±2°C, aire filtrado, iluminación con lámparas blancas 20h luz/4h oscuridad, irradianza 70 μE m-2s-1, diferentes concentraciones de acetato de sodio y cloruro de sodio. Se determinó crecimiento celular, cambios morfológicos y cuantificación de astaxantina y clorofila por espectrofotometría. Se realizó un análisis estadístico utilizando ANOVA (95%). Resultados. Utilizando 0,299 mg/L de acetato de sodio se obtuvo un crecimiento celular de 2,0 x 104 Cel/mL y una concentración de astaxantina de 2,530 μg/mL, mientras que con 1,6 mg/L de acetato de sodio el crecimiento celular fue de 3,5 x 104 Cel/mL y una concentración de astaxantina de 1,9 μg/ml. El tratamiento al cual se le adicionó 1,6 g/L de acetato de sodio y 6,4 g/L de cloruro de sodio presentó la mayor producción astaxantina 7,3 μg/ml. El tratamiento con acetato de sodio 0,320 g/L + cloruro de sodio 1,28 g/L presentó el mayor crecimiento celular con 1,64x105 células/ml. Conclusión. Esta investigación destaca la importancia de cultivar inicialmente la microalga utilizando el biorreactor Tecferm de 5 litros y después de su fase exponencial someterla a factores de estrés con acetato de sodio y cloruro de sodio lográndose así la mayor producción de astaxantina 7,325 μg/ml.


Abstract Introduction. Haematococcuspluvialis is a microalgae that produces astaxanthin, a beta-carotene and antioxidant widely used in industry. In order to obtain a higher production of astaxanthin, the objective was to use different stress factors, in a 5-liter laboratory-scale bioreactor. Methodology. The microalgae was cultivated in the RM medium, pH 6.8, temperature 20 ± 2°C, filtered air, illumination with white lamps 20h light/4h darkness, irradiance 70 μE m-2s-1, different concentrations of sodium acetate and chloride of sodium. Cell growth, morphological changes and quantification of astaxanthin and chlorophyll were determined by spectrophotometry. Statistical analysis was performed using ANOVA (95%). Results. Using 0.299 mg/L of sodium acetate a cell growth of 2.0 x 104 Cel/mL and an astaxanthin concentration of 2.530 μg/mL were obtained, while with 1.6 mg/L of sodium acetate the cell growth It was 3.5 x 104 Cel/mL and an astaxanthin concentration of 1.9 μg/mL. The treatment to which 1.6 g L of sodium acetate and 6.4 g/L of sodium chloride were added showed the highest astaxanthin production, 7.3 μg/ml. Treatment with 0.320 g/L sodium acetate + 1.28 g/L sodium chloride showed the highest cell growth with 1.64x105 cells/ml. Conclusion. This research highlights the importance of initially cultivating the microalgae using the 5-liter Tecferm bioreactor and, after its exponential phase, subjecting it to stress factors with sodium acetate and sodium chloride, thus achieving the highest production of 7.325 μg/ml astaxanthin.

ABSTRACT

Helicobacter pylori is a dominant bacterium living in the human gastric tissues. In H. pylori-infected tissues, the infiltrated inflammatory cells produce reactive oxygen species (ROS), leading to gastric inflammation with production of various mediators. According to numerous epidemiological studies, dietary carotenoids may prevent gastric inflammation due to their antioxidant properties. Recent studies showed that antioxidant and anti-inflammatory effects of astaxanthin and β-carotene may contribute to inhibition of H. pylori-induced gastric inflammation. Astaxanthin changes H. pylori-induced activation of T helper cell type 1 response towards T helper cell type 2 response in the infected tissues. Astaxanthin inhibits the growth of H. pylori. Even though astaxanthin reduces H. pylori-induced gastric inflammation, it does not reduce cytokine levels in the infected tissues. β-Carotene suppresses ROS-mediated inflammatory signaling, including mitogen-activated protein kinases and redox-sensitive transcription factors, and reduces expression of inflammatory mediators, including interleukin-8, inducible nitric oxide synthase, and cyclooxygenase-2 in the infected tissues. Therefore, consumption of astaxanthin- and β-carotene-rich foods may be beneficial to prevent H. pylori-induced gastric inflammation. This review will summarize anti-inflammatory mechanisms of astaxanthin and β-carotene in H. pylori-mediated gastric inflammation.

ABSTRACT

Background: Introgression of transgenes from crops to their wild species may enhance the adaptive advantage and therefore the invasiveness of and weedy forms. The study evaluated the effect of Africa Biofortified Sorghum (ABS) genes from ABS event 188 on the vegetative and reproductive features of the F2 populations derived from crosses with Sorghum bicolor subsp. drummondii. Results: F1 populations were obtained from reciprocal crosses involving ABS event 188 and its null segregant with inbred weedy parents from S. bicolor subsp. drummondii. Four F2 populations and four parental populations were raised in RCBD with 4 replications in a confined field plot for two seasons. Vegetative and reproductive traits were evaluated. The vigour shown in the F2 populations from the reciprocal crosses involving ABS event 188 and S. bicolor subsp. drummondii was similar to that in the crosses involving the null segregant and S. bicolor subsp. drummondii. Differences in vegetative and reproductive parameters were observed between the parental controls and the F2 populations. Examination of the above and below ground vegetative biomass showed lack of novel weedy related features like rhizomes. Conclusions: Therefore, release of crops with ABS 188 transgenes into cropping systems is not likely to pose a risk of conferring additional adaptive advantage in the introgressing populations. The interaction of ABS genes in weedy backgrounds will also not have an effect towards enhancing the weedy features in these populations.

ABSTRACT

Beta-carotene is a provitamin, i.e. a precursor of vitamin A (retinol), which is classified as an essential nutrient for humans. Beta-carotene is one of many carotenoids found in plants, fungi and bacteria. Carotenoids are therefore predominantly obtained through foods of plant origin or food supplements. Carrots contribute approximately half of the total beta-carotene intake in the Norwegian diet, followed by mixed frozen vegetables, tomatoes, fruits and berries. VKM emphasises that this opinion on upper level (UL) for beta-carotene addresses beta-carotene in food supplements only. Beta-carotene from regular foods such as vegetables and fruits is not considered to be a health concern. In 2002, the Scientific Committee on Food (SCF) established a tolerable upper intake level (UL) for vitamin A (SCF, 2002). However, the SCF opinion covers only retinol compounds (various forms of vitamin A). The bioconversion of carotenoids to vitamin A in the body is well regulated and therefore only intake of vitamin A has been considered relevant for vitamin A toxicity (Blomhoff et al., 2003; EFSA, 2008). The Norwegian Food Safety Authority is considering whether betacarotene should be regulated separately from retinol compounds. Beta-carotene seems to have a carcinogenic effect in smokers. A number of studies have been published where possible mechanisms of this negative health effect are discussed. The suggested mechanisms are either related to effects on cytochrome P450-related activities, altered retinoid signalling or to a pro-oxidant activity of beta-carotene. No UL has been established for beta-carotene. Several risk-assessment bodies have, however, previously attempted to establish safe levels or temporary guidelines, summarised in the following table: Previous reports Conclusion SCF, 2000, EU No dose-response relationship could be derived. Supplementation of 20 mg beta-carotene per day or more is contraindicated for use in current heavy smokers. There is insufficient evidence to set an UL for beta-carotene. IOM, 2000, USA No UL was established for beta-carotene or carotenoids. Beta-carotene supplementation is not recommended in the general population. EVM, 2003, UK The LOAEL was set to 20 mg/day. An uncertainty factor of 3 was applied to extrapolate from LOAEL to a NOAEL. A Safe Upper Level for beta-carotene supplements was set at 7 mg/day (equivalent to 0.11 mg/kg body weight/day for a 60 kg adult). NNR, 2012, Nordic countries No specific beta-carotene recommendation or UL. Rasmussen, 2006, Denmark A Temporary Guidance Level for beta-carotene equal to the average dietary level of 5 mg/day for all age groups was suggested. Seven randomised controlled trials (RCTs) have been included in this VKM opinion, conducted either in Europe or the USA, with almost 47 100 participants in the beta-carotene groups. In six of these RCTs there were no observed increased risk of cancer, but the large Finnish ATBC study found an increased risk of lung cancer in the beta-carotene group. Two prospective studies were included, one Danish and one from the USA, with 125 000 participants all together. The Danish study found that risk of lung cancer increased in smokers with increasing doses of beta-carotene supplements. In addition, eleven meta-analyses were included; one with age-related macular degeneration as endpoint, one with a mixture of cardiovascular disease (CVD) and cancer as endpoints, four on cancer as only endpoint, one on a mixture of CVD and all-cause mortality as endpoints and four on all-cause mortality alone. One of the meta-analyses on all-cause mortality was later excluded. There were no significant findings in the meta-analysis on macula degeneration. One of the two meta-analyses on CVD found a small increased risk in the beta-carotene arm (Vivekananthan et al., 2003). The combined CVD and cancer meta-analysis did not have sufficient statistical power to get significant results, but found a probable increase in lung cancer incidence in high-risk subgroups (smokers and asbestos workers (Fortmann et al., 2013). In the five meta-analyses studying cancer, there were no effect on other cancer forms than lung cancer. The four meta-analyses on all-cause mortality used information from the same RCTs as included in this VKM-opinion. They extracted information on numbers of death in each study and used these numbers to analyse risk of death in the beta-carotene versus placebo groups. Alarmingly, they all found an increased risk of all-cause mortality. These meta-analyses have been discussed thoroughly. To complete the risk characterisation of beta-carotene, VKM has followed the steps 1 – 4 as suggested by SCF in their Guidelines for the development of tolerable upper intake levels for vitamins and minerals (SCF, 2000a). Step 1 and 2. Hazard identification and characterisation Up until two decades ago, beta-carotene was thought to be harmless even in large doses. In the wake of the Finnish ATBC study which found an increased risk of lung cancer and death in male smokers, animal studies have indicated three possible mechanisms for such a detrimental effect. Although conclusive mechanistic explanations for the negative effects have not yet been agreed upon, there is a scientific rationale for the argument that population groups with vulnerable lungs may also have increased risk from beta-carotene supplements. The dose used in the Finnish ATBC study was 20 mg beta-carotene/day. The effect was only observed during the intervention period; in follow-up studies conducted after the active period was finished, the risk declined and was no longer significant. 20 mg beta-carotene may thus be considered as a LOAEL. The Danish prospective study found a dose-dependent increase in lung cancer risk with increased intake of supplemental beta-carotene. Unfortunately, the paper does not allow for setting a NOAEL or LOAEL based on the published data. In the four meta-analyses on all-cause mortality, all found a 6-7% increased risk of death. One of the meta-analyses also found an increased risk of CVD in smokers. However, all results were driven, statistically, by the ATBC study. Studies with a more mixed population (both men and women) and with a more typical prevalence of smokers (10–20%), found no such increased risk. Step 2, continued: Derive at a UL, taking into account the scientific uncertainties in the data. ULs may be derived for various life-stage groups within the population VKM found it extraordinary challenging to decide which uncertainty factor to use for beta-carotene. The present SCF guidelines for establishment of tolerable upper intake levels do not give clear guidance/advice in deciding the numeric level of the uncertainty factor. This seems to leave the decision to scientific judgement. If the NOAEL is based on human data, an uncertainty factor of 10 is recommended as a starting point to encompass inter-individual variation and sensitivity. The SCF guidelines state that a small uncertainty factor is to be used if the judgement is that little population variability is expected for the adverse effects, and a larger uncertainty factor (close to 10) may be used if variability is expected to be large. For beta-carotene, a NOAEL is not available, and an uncertainty factor may be applied to account for the uncertainty in deriving a UL from the LOAEL. The size of the uncertainty factor involves a judgement based on the severity and incidence of the observed effect at the LOAEL and the steepness (slope) of the dose response, if this is possible to estimate. For beta-carotene, we have not found the data necessary to make a dose-response curve. In addition, the following considerations were discussed before deciding on an uncertainty factor: · The study which found a negative effect of beta-carotene supplementation (the Finnish ATBC study) was very large (n=29 133) which indicates that it encompasses inter-individual variation and sensitivity. Additionally, the most vulnerable groups, in this case smokers, was an inclusion criteria. Both these factors indicate that the uncertainty factor can be in the lower end. · The meta-analyses for the endpoint “increased risk of all-cause mortality” found an increased risk of death in the beta-carotene groups. This is severe, and indicates that a maximum uncertainty factor should be applied. However, as all results in the all-cause mortality metaanalyses were driven statistically, by the ATBC study on smokers, we choose to use a lower factor. Based on the above considerations, VKM has chosen to use 5 as an uncertainty factor for betacarotene. An UL for beta-carotene cannot be derived, but a tentative upper level (TUL) is set at 4 mg/day, based on a LOAEL of 20 mg and the uncertainty factor of 5. Smokers and anyone else in the population with vulnerable lungs (e.g. asthmatics, COPD patients) should be discouraged from taking beta-carotene containing supplements all together. Step 3. Exposure assessment – evaluates the distribution of usual total daily nutrient intakes among members of the general population In the food survey Småbarnskost 2007, the mean intake of beta-carotene in 2-year-olds was 1.5 mg/day. In Norkost 3, the estimated mean intake in adults was 2.4 mg/day and 6.9 mg/day in the 95th percentile. About 3% of the adults reported use of beta-carotene supplements. The use of tanning pills containing beta-carotene may have been underreported. Beta-carotene from regular foods such as vegetables and fruits is, however, not considered to be of any health concern. Negative health effects from beta-carotene in natural foods have never been reported. On the contrary, the consumption of vegetables and fruits should be increased, and the recommendation of “5 a day” should be achieved in all age groups of the population. Step 4. Risk characterisation – analyses of the conclusions from steps 1 through 3 and characterises the risk. The risk will depend on the fraction of the population

Los carotenoides son pigmentos isoprenoides, que están presentes de manera natural y en altas concentraciones en la mayoría de alimentos vegetales, por ejemplo, aguacate (palta), papaya, tomate y zanahoria, pero también se encuentran como aditivos y colorantes en diversos productos cárnicos, aceites vegetales, salsas, aderezos, harinas, entre otros. En los últimos años, varios grupos de investigación han descrito que los carotenoides de la dieta participan en varios procesos fisiopatológicos, incluyendo, la respuesta inmune de tipo alérgico. Las tasas de prevalencia para este tipo de enfermedades se han incrementado de manera alarmante durante las últimas cinco décadas en todo el mundo, y varios factores e hipótesis se han planteado tratando de explicar este fenómeno. Uno de ellos es la hipótesis de la dieta, la cual plantea que la composición de varios micro- y macronutrientes de la alimentación, así como los cambios en los patrones dietarios, serían los responsables de dicho fenómeno. En adición, evidencia epidemiológica y experimental señala que los carotenoides de la dieta participan en la regulación de la inflamación alérgica y, por ello, se postulan como coadyuvantes en la terapia de estos padecimientos. En el presente manuscrito se revisará el estado del arte en relación con los efectos de los carotenoides de la dieta sobre el estado inflamatorio alérgico.(AU)


The carotenoids are isoprenoids pigments, which are naturally present in high concentrations and in most vegetables foods e.g., avocado, papaya, tomato and carrot, but also as additives and dyes found in various meat products, vegetable oils, sauces, dressings, flour, among others. In recent years, several research groups have reported that dietary carotenoids participate in many pathophysiological processes, including the allergic immune response. The prevalence rates for these diseases have increased alarmingly over the past five decades in the world and a number of factors and assumptions have been proposed trying to explain this phenomenon, one of them is the hypothesis of the diet, which has arisen that the composition of several micro and macronutrients of foods, as well as changes in the dietary patterns would be responsible for thisphenomenon.In addition, epidemiological and experimental evidence indicates that dietary carotenoids are involved in the regulation of allergic inflammation and thus postulated as adjuvants in the therapy of these diseases. In this manuscript, we will review the state of art in relation to the effects of dietary carotenoids on the allergic inflammatory state.(AU)

Vitamin A, C and E intake has been shown to play a role in the etiology of breast cancer, but the findings have been inconsistent and limited to developed countries with higher cancer incidence. Therefore, the aim of this study is to examine the association of premenopausal and postmenopausal breast cancer risk with vitamin A, C and E intake from dietary sources. This is a population based case-control study conducted in Malaysian population among 382 breast cancer patients and 382 control group. Dietary intake was assessed via an interviewer-administered food frequency questionnaire. Logistic regression was used to compute odds ratios (OR) with 95% confidence intervals (CI) and a broad range of potential confounders were included in analysis. The results of this study shows a significant decreased risk of breast cancer among premenopausal (ORQ4 to Q1=0.38, 95% CI, 0.12 – 0.55, p-trend=0.001) and postmenopausal (ORQ4 to Q1=0.26, 95% CI, 0.03 – 0.75, p-trend=0.017) women was observed in the highest quartile of beta-carotene intake. Meanwhile, a higher intake of vitamin C showed significantly lowered risk only for premenopausal women (ORQ4 to Q1=0.13, 95% CI, 0.03 – 0.32, p-trend=0.001). As a conclusion, beta-carotene intake was independently related to pre- and postmenopausal breast cancer risk, while vitamin C intake was associated with decreased risk among premenopausal women only. However, no association was observed for vitamin A especially retinol and vitamin E intake from dietary sources.

ABSTRACT

Objective: to investigate the effect of administration of supraphysiological âcaroteno on biological parameters (ectoscopy and blood pressure), laboratory (malondialdehyde) and histological (liver and carotid arteries) of spontaneously hypertensive rats prone to stroke (SHR-sp). Methods: we used 36 male rats were divided into three groups, each containing 12 rats Wistar, SHR and SHR-sp, subdivided into six control animals and six animals treated with supraphysiological doses of âcaroteno for two periods of ten weeks interspersed with one week interruption. In the experiment were assessed daily physical examination and blood pressure (plethysmography). At sacrifice, blood was collected for measurement of serum malondialdehyde, liver and carotid arteries for histological examination. Results: temporary change in color of the fur, decreased significantly (p<0.0001) blood pressure (20mg supplementation âcaroteno) and serum levels of malondialdehyde (p<0.05) and increased amount of elastic fibers in the carotid wall of SHR and SHR-sp. Conclusion: supplementation of supraphysiological âcaroteno caused no toxic effects, showed positive response in the modulation of blood pressure and lower serum malondialdehyde. No significant morphological changes were found in both groups, except an increase in the number of elastic fibers in the muscle layer carotid suggesting elastosis in SHR and SHR-sp. .


Objetivo: investigar o efeito da administração suprafisiológica de bcaroteno sobre parâmetros biológicos, laboratoriais e histológicos dos ratos espontaneamente hipertensos com tendência ao acidente vascular encefálico (SHR-sp). Métodos: utilizaram-se 36 ratos machos, distribuídos em três grupos, contendo cada um dos 12 ratos das linhagens Wistar, SHR e SHR-sp, subdivididos em seis animais controle e seis animais tratados com doses suprafisiológicas de âcaroteno por dois períodos de dez semanas, intercalados por uma semana de interrupção. No experimento foram avaliados diariamente o exame físico e a pressão arterial. Foi coletado sangue para dosagem sérica de malondialdeído; o fígado e as artérias carótidas para exame histológico. Resultados: alteração provisória na coloração dos pelos, diminuição significativa (p<0,0001) da pressão arterial (suplementação de 20mg de bcaroteno) e dos níveis séricos de malondialdeído (p<0,05) e aumento da quantidade de fibras elásticas na parede carotídea dos ratos SHR e SHR-sp. Conclusão: A suplementação suprafisiológica de bcaroteno não causou efeitos tóxicos, apresentou resposta positiva na modulação da pressão arterial e diminuição na concentração sérica de malondialdeído. Não foram encontradas alterações morfológicas significativas nos grupos estudados, exceto um aumento no número de fibras elásticas da camada muscular carotídea sugerindo elastose nos ratos SHR e SHR-sp. .

ABSTRACT

Dentre as espécies cultivadas no semiárido baiano, as abóboras são uma excelente alternativa de produção. Nessa região onde são encontrados os mais baixos Índices de Desenvolvimento Humano (IDH) do Brasil e populações com alta incidência de hipovitaminose A, materiais promissores de abóboras, fontes de carotenoides, podem ser inseridos no agronegócio irrigado. O objetivo do trabalho foi identificar acessos de abóboras do Banco Ativo de Germoplasma (BAG) de Cucurbitáceas da Embrapa Semiárido com altos teores de carotenoides totais e vitamina A. Os teores de carotenoides totais (CT), expressos em termos de β-caroteno, variaram de 14,93µg g-1 (A4) até 290,62µg g-1 (A43) entre os diferentes acessos (A). Dos carotenoides encontrados nos acessos com maiores teores de CT, 60-70% foram quantificados como α- e β-caroteno, representando um valor total de vitamina A de 1.182,39 gRAE 100g-1 (A40) e 1.828,84 gRAE 100g-1 (A43). Esses resultados demonstram que os 48 acessos de abóboras do BAG estudados apresentam grande variação quanto ao teor de carotenoides totais, e os selecionados representam alta fonte de vitamina A.


Among the species of short cycle crops, with great potential to be increased within the agribusiness of the Semi-Arid Bahia, the pumpkins are an excellent alternative for production. In this region where are found the lowest HDI of Brazil, and populations with high incidence of hypovitaminosis A, promising materials of pumpkins with high amounts of carotenoids have a chance to be good accepted and inserted into the irrigated agribusiness. The aim of this research was to identify accessions of pumpkins from the Active Germplasm Bank (BAG) of Cucurbitáceas from Embrapa Semi-arid with high amounts of total carotenoids and vitamin A. The amount of total carotenoids found by UV-vis spectrophotometry (445nm) in different accessions (A) ranged from 14.93mg g-1 (A4) to 290.62mg g-1 (A43), with average value of 366.60 31.21µg g-1. In the access with higher amounts of total carotenoids a percentage between 60-70 % were quantified as α- and β-carotene showing a total value of vitamin A of 1,182.39 gRAE 100g-1 (A40) and 1,828.84 gRAE 100g-1 (A43). These results demonstrate that the 48 accessions of pumpkins BAG studied show a big variation in the content of total carotenoids, and may represent some high source of vitamin A, which really increase the importance of identifying genotypes for production in large scale of lines segregant of vitamin A.

ABSTRACT

Atualmente, são produzidas milhões de toneladas de resíduos provenientes do processamento agroindustrial. Muitos deles são ricos em compostos bioativos sendo potenciais fontes naturais dessas substâncias. Assim, este trabalho teve como objetivo avaliar o teor de compostos fenólicos totais, a atividade antioxidante e a composição fenólica de três resíduos gerados por agroindústrias brasileiras: bagaço de uva Isabel (BI) (Vitis labrusca), bagaço de uva Verdejo (BV) (Vitis vinifera) e bagaço de goiaba (BG) (Psidium guajava). Os resultados do teor de compostos fenólicos totais (mg GAE g-1) encontrados nos extratos etanólicos e aquosos dos resíduos foram, respectivamente: BV (20,94±0,46; 8,03±0,43)> BI (16,57±0,19; 4,41±0,01)> BG (3,41±0,09; 1,88±0,06). Alta atividade antioxidante, principalmente em BV e BI, foi verificada nos ensaios realizados (ABTS ●, DPPH ● e auto-oxidação do sistema beta-caroteno/ácido linoléico). Uma forte correlação positiva entre atividade antioxidante e o teor de compostos fenólicos totais foi encontrada. Os compostos fenólicos encontrados, por cromatografia gasosa com espectrometria de massas (CG-EM), foram: ácido gálico, epicatequina, quercetina (BV, BI e BG); ácido isovanílico (BI, BG); ácido p-cumárico (BI); ácido caféico e resveratrol (BV, BI). Esses resultados mostram que os resíduos agroindustriais analisados, particularmente os vinícolas, são ricos em substâncias bioativas e podem ser explorados pela indústria de alimentos e farmacêutica.


Nowadays, the agro-industrial processing produces millions of tons of wastes. Many of them are rich in bioactive compounds, being a potential natural source of these substances. This study aimed to evaluate the content of total phenolics, antioxidant activity and phenolic composition of residues generated by three Brazilian agribusiness: Isabel grape pomace (PI) (Vitis labrusca), Verdejo grape pomace (PV) (Vitis vinifera) and guava pomace (PG) (Psidium guajava). The results of total phenolics content (mg GAE g-1) found in the ethanol and aqueous extracts of residues were: PV (20.94±0.46; 8.03±0.43)>PI (16.57±0.19; 4.41±0.01)>PG (3.41±0.09; 1.88±0.06). High antioxidant activity of these extracts, particularly PV and PI, was found by the methods DPPH ●, ABTS ● and beta-carotene bleaching method. A strong positive correlation between antioxidant activity and content of total phenolic compounds was found. The following phenolic compounds were found by gas chromatography with mass spectrometry (GC-MS): gallic acid, epicatechin, quercetin (PV, PI and PG); isovanilic acid (PI, PG), p-coumaric acid (PI), caffeic acid and resveratrol (PV, PI). The results show that these residues, particularly the wineries, are rich in bioactive substances and should be exploited by the food industry and pharmaceuticals.

ABSTRACT

High consumption of fruits and vegetables has been suggested to provide some protection to smokers who are exposed to an increased risk of numerous cancers and other degenerative diseases. Carrot is the most important source of dietary beta-carotene. Therefore, the objective of this study was to investigate whether carrot juice supplementation to smokers can protect against lymphocyte DNA damage and to compare the effect of supplementation of capsules containing purified beta-carotene or a placebo (simple lactose). The study was conducted in a randomized and placebo-controlled design. After a depletion period of 14 days, 48 smokers were supplemented with either carrot juice (n = 18), purified beta-carotene (n = 16) or placebo (n = 14). Each group was supplemented for 8 weeks with approximately 20.49 mg of beta-carotene/day and 1.2 mg of vitamin C/day, as carrot juice (300 ml/day) or purified beta-carotene (20.49 mg of beta-carotene, 1 capsule/day). Lymphocyte DNA damage was determined using the COMET assay under alkaline conditions and damage was quantified by measuring tail moment (TM), tail length (TL), and% DNA in the tail. Lymphocyte DNA damage was significantly decreased in the carrot juice group in all three measurements. The group that received purified beta-carotene also showed a significant decrease in lymphocyte DNA damage in all three measurements. However, no significant changes in DNA damage was observed for the placebo group except TM (P = 0.016). Erythrocyte antioxidant enzyme was not significantly changed after supplementation. Similarly plasma lipid profiles were not different after carrot juice, beta-carotene and placebo supplementation. These results suggest that while the placebo group failed to show any protective effect, carrot juice containing beta-carotene or purified beta-carotene itself had great antioxidative potential in preventing damage to lymphocyte DNA in smokers.

As deficiências de vitaminas, verificadas em indivíduos com HIV/Aids em terapia antirretroviral de alta potência (HAART) têm sido associadas à piora do curso clínico da doença e maior risco de mortalidade. Objetivo Mostrar a distribuição das concentrações séricas de vitaminas A e E, e beta-caroteno em adultos com infecção pelo HIV/Aids e estudar a associação de suas concentrações, segundo diferentes esquemas de HAART. Métodos Foram selecionados 182 adultos de 20 a 59 anos de idade, de ambos os sexos, com HIV/Aids em HAART estável por no mínimo 6 meses, e com níveis de linfócitos T-CD4+ 200 células/mm3. Os indivíduos foram divididos em três grupos por esquema de HAART utilizado: inibidores de transcriptase reversa análogos de nucleosídeo (ITRN) associados a inibidores de transcriptase reversa não análogos de nucleosídeo (ITRNN); ITRN associada a inibidores de protease (IP); ITRN associadas a outras classes (inibidores de fusão, inibidores de integrasse, inibidores de entrada e IP associada a essas medicações). A determinação dos micronutrientes foi realizada por cromatografia líquida de alta eficiência. Foram verificadas variáveis sócio-demográficas e econômicas, estilo de vida, história da doença, uso de medicações e variáveis antropométricas e laboratoriais. Para medir os efeitos das variáveis explanatórias (esquemas de tratamento, tempo de uso e adesão ao último esquema) sobre cada variável resposta (retinol, alfa-tocoferol e beta-caroteno), foram realizadas análises de regressão linear múltipla. Sexo, idade, escolaridade, tabagismo, prática de atividade física, tempo de infecção por HIV, presença de comorbidades, relação cintura-quadril e níveis de linfócitos T-CD4+ e colesterol foram usadas como variáveis de controle. Deficiency of vitamins found in individuals with HIV/AIDS on highly active antiretroviral therapy (HAART) has been associated with an increased risk of disease progression and mortality. Objective To show the distribution of serum concentrations of vitamins A and E and beta-carotene in adults with HIV/AIDS, and to study the association of their concentrations, according to different regimens of HAART. Methods We selected 182 men and women aged 20-59 years with HIV/AIDS on stable HAART for at least six months and with levels of CD4+ T-lymphocytes 200 cells/mm3. Individuals were divided into three groups according to the HAART regimen used: nucleoside reverse transcriptase inhibitors (NRTI) combined with nonnucleoside reverse transcriptase inhibitors (NNRTI); NRTI combined with protease inhibitors (PI); NRTI combined with other classes (fusion inhibitors, integrase inhibitors, entry inhibitors, and PI associated with these medications). Determinations of vitamins A and E and beta-carotene were performed by high-performance liquid chromatography. Socio-demographic and economic variables, lifestyle, disease history, medication use, and anthropometric and laboratory variables were assessed. Multiple regression analyses were used to measure the effects of the explanatory variables (treatment regimens, duration and adherence to the last treatment regimen) on each response variable (retinol, alpha-tocopherol and beta-carotene). ABSTRACT

Elevated plasma concentration of total homocysteine (ptHcy) is known as an independent risk factor of cardiovascular disease (CVD) and oxidative stress is also commonly implicated in CVD. An association between ptHcy and oxidative stress has recently been suggested. The study objective is to examine the relationship between ptHcy and oxidative stress markers in 103 healthy college students (62 males and 41 females). Plasma levels of ptHcy, oxidative stress markers (conjugated diene, erythrocyte catalase, TRAP, lymphocyte DNA damage), antioxidant vitamins (alpha-tocopherol, gamma-tocopherol, carotenoids), and lipid parameters (total cholesterol, triglyceride, HDL cholesterol) were determined. The results show that the concentration of ptHcy was significantly higher in male subjects (22.17 +/- 2.14 micromole/L) than in female subjects (12.28 +/- 0.45 micromole/L). There was a negative association between ptHcy and plasma beta-carotene in male subjects (p or = 15 micromol/L), as compared to those with lower plasma homocysteine. These study results confirmed the views on the association between plasma homocysteine and oxidative stress markers in humans and support the hypothesis that homocysteine promotes the oxidative environment by counteracting the antioxidant defense mechanism.

ABSTRACT

PURPOSE: Elevated circulating oxidized low density lipoprotein (Ox-LDL) levels are associated with increased risk of atherosclerosis, which may be due to high plasma homocysteine (Hcy) and low intakes of antioxidants. We investigated the contribution of dietary intakes of antioxidants to Hcy-induced LDL oxidation in atherosclerotic patients (AP) and controls. MATERIALS AND METHODS: Male AP (n = 101) who were confirmed by coronary angiography and 91 controls were evaluated by blood biochemistry and dietary intakes. To determine whether homocysteine is an independent risk factor for atherosclerosis, subjects were divided into three groups; low- ( or = 12.1 uM/L) Hcy. RESULTS: Plasm levels of homocysteine and LDL were higher, but plasma apo A-I in HDL and folate were lower in the AP group. The odds ratio (OR) for the risk of atherosclerosis was 3.002 [95% confidence interval (CI), 1.27-7.09] for patients in the highest tertile with homocysteine > or = 12.1 uM/L. AP having high homocysteine levels had low intakes of vitamin A, beta-carotene and vitamin C. By logistic regression analysis, age, body mass index (BMI), plasma LDL, plasma folate, and low intakes of vitamin A and beta-carotene were found to be risk factors for atherosclerosis in patients with high-Hcy, but dietary B vitamins including folate were not. CONCLUSION: A high-Hcy level was a risk factor for atherosclerosis in patients with high Ox-LDL levels. High intakes of antioxidants appeared to be a protective factor for atherosclerosis, perhaps exerting a pro-oxidative effect on LDL when combined with high levels of Hcy and LDL. However, more evidence for the benefits of B vitamins as a homocysteine-lowering therapy is needed.

FUNDAMENTO: Os mecanismos envolvidos na maior remodelação causada pelo betacaroteno após o infarto são desconhecidos. OBJETIVO: Analisar o papel da lipoperoxidação na remodelação ventricular após o infarto do miocárdio, em ratos suplementados com betacaroteno. MÉTODOS: Ratos foram infartados e distribuídos em dois grupos: C (controle) e BC (500mg/kg/dieta). Após seis meses, foram realizados ecocardiograma e avaliação bioquímica. Utilizamos o teste t, com significância de 5 por cento. RESULTADOS: Os animais do grupo BC apresentaram maiores médias das áreas diastólicas (C = 1,57 ± 0,4 mm²/g, BC = 2,09 ± 0,3 mm²/g; p < 0,001) e sistólicas (C = 1,05 ± 0,3 mm²/g, BC = 1,61 ± 0,3 mm²/g; p < 0,001) do VE, ajustadas ao peso corporal do rato. A função sistólica do VE, avaliada pela fração de variação de área, foi menor nos animais suplementados com betacaroteno (C = 31,9 ± 9,3 por cento, BC = 23,6 ± 5,1 por cento; p = 0,006). Os animais suplementados com betacaroteno apresentaram valores maiores da relação E/A (C = 2,7 ± 2,5, BC = 5,1 ± 2,8; p = 0,036). Não foram encontradas diferenças entre os grupos em relação aos níveis cardíacos de GSH (C = 21 ± 8 nmol/mg de proteína, BC = 37 ±15 nmol/mg de proteína; p = 0,086), GSSG (C = 0,4 (0,3-0,5) nmol/g de proteína, BC = 0,8 (0,4-1,0; p = 0,19) de proteína; p = 0,246) e lipoperóxidos (C = 0,4 ± 0,2 nmol/mg de tecido, BC = 0,2 ± 0,1 nmol/mg de tecido; p = 0,086). CONCLUSÃO: A maior remodelação em animais infartados e suplementados com betacaroteno não depende da lipoperoxidação.


BACKGROUND: The mechanisms involved in the biggest remodeling caused by the post-infarct beta-carotene are unknown. OBJECTIVE: To analyze the role of lipoperoxidation in the ventricular remodeling after infarct of the myocardium in rats supplemented with beta-carotene. METHODS: Rats were infarcted and divided into two groups: C (control) and BC (500mg/kg/regimen). After six months, echocardiogram and biochemical evaluation were performed. The t test was used, with 5 percent significance. RESULTS: The animals from BC group presented highest means of the diastolic (C = 1.57 ± 0.4 mm²/g, BC = 2.09 ± 0.3 mm²/g; p < 0.001) and systolic (C = 1.05 ± 0.3 mm²/g, BC = 1.61 ± 0.3 mm²/g; p < 0.001) areas of LV, which were adapted according to the rat's body weight. The systolic function of LV, evaluated by the area variation fraction, was lower in the animals supplemented with beta-carotene (C = 31.9 ± 9.3 percent, BC = 23.6 ± 5.1 percent; p = 0.006). The animals supplemented with beta-carotene presented higher values of the E/A relation (C = 2.7 ± 2.5, BC = 5.1 ± 2.8; p = 0.036). No differences were found between the groups concerning the cardiac levels of the GSH (C = 21 ± 8 nmol/mg of protein, BC = 37 ± 15 nmol/mg of protein; p = 0.086), GSSG (C = 0.4 (0.3-0.5) nmol/g of protein, BC = 0.8 (0.4-1.0; p = 0.19) of protein; p = 0.246) and lipoperoxides (C = 0.4 ± 0.2 nmol/mg of tissue, BC = 0.2 ± 0.1 nmol/mg of tissue; p = 0.086). CONCLUSION: The highest remodeling in infarcted rats supplemented with beta-carotene does not depend on the lipoperoxidation.

RESULTADOS: Los animales del grupo BC presentaron mayores promedios de las áreas diastólicas (C = 1,57 ± 0,4 mm²/g, BC = 2,09 ± 0,3 mm²/g; p < 0,001) y sistólicas (C = 1,05 ± 0,3 mm²/g, BC = 1,61 ± 0,3 mm²/g; p < 0,001) del VI, ajustadas al peso corporal de la rata. Smoking is associated with an increased incidence of numerous cancers and other degenerative diseases. It has been suggested that high consumption of fruits and vegetables may give some protection. Especially carrot is the most important source of dietary beta-carotene. Therefore, the objective of this study is to investigate whether carrot juice supplementation to smokers have different or superior effect of compared to the effect supplementing purified beta-carotene. The study was conducted in a randomized and placebo-controlled design, after a depletion period of 14 days, 48 smokers were supplemented either carrot juice (n = 18), purified beta-carotene (n = 16) or placebo (n = 14). Each group was supplemented for 8 weeks with approximately 20.49 mg of beta-carotene/day and 1.2 mg of vitamin C/day, as carrot juice (300 mL/day) or purified beta-carotene (1 capsule/day). Plasma vitamin C, vitamin E and beta-carotene level were significantly increased after carrot juice and beta-carotene supplementation. These results suggest that carrot juice containing beta-carotene or beta-carotene itself have similar antioxidative potentials by increasing the antioxidant potential in smokers. Therefore, we suggest moderate dose of vitamin supplementation (amount of two servings of vegetable intake) may help to replenish the decreased oxidative stress levels in smokers.

ABSTRACT

Objetivou-se avaliar procedimentos de manipulação visando o controle de perdas de β-caroteno e licopeno em couve e tomate preparados em uma Unidade de Alimentação e Nutrição (UAN) hospitalar. Os critérios adotados não utilizados previamente pela UAN foram: armazenamento por 24 h sob refrigeração (10°C), sanitização por 15 min e distribuição logo após o preparo. As hortaliças foram coletadas após a recepção e depois de cada etapa de manipulação. A análise foi realizada por cromatografia líquida de alta eficiência (CLAE), usando como fase móvel metanol, acetato de etila e acetonitrila (50:40:10). A ANOVA (α = 0,05) foi utilizada para detecção de diferenças significativas. Não foram encontradas diferenças significativas quanto ao conteúdo dos componentes entre as etapas de manipulação, mas houve redução importante das taxas de retenção. Para β-caroteno em couve, verificou-se retenção de 68,2 por cento após 60 minutos de exposição para consumo, enquanto em tomate, 91,96 por cento do conteúdo desse composto foi preservado após 120 minutos de espera até a distribuição. Não foi observada redução importante na taxa de retenção de licopeno. Os procedimentos avaliados na UAN hospitalar contribuíram para controlar as perdas de carotenóides nas hortaliças, pois sua retenção foi elevada, sendo sugerida sua adoção em outras UAN.


The aim of this study was to assess the handling procedures of kale and tomatoes in a hospital foodservice (HFS) in order to control loss of β-carotene and licopene. The adopted measures, up to then not used by the HFS, were: 24-h storage under refrigeration (10°C), hygienizing for 15 min and distribution immediately after preparation. Vegetable samples were collected after reception and after each stage of manipulation in the HFS. The samples were analyzed using high performance liquid chromatography (HPLC) with a mobile phase of methanol, ethyl acetate and acetonitrile (50:40:10). ANOVA (α = 0.05) was used to detect significant differences. No significant differences in the content of the components were found between the different stages of manipulation but there was an important decrease in the retention rates. In kale a retention rate of 68.2 percent for β-carotene was verified 60 minutes after preparation whereas in tomatoes 91.96 percent of this compound were preserved for 120 minutes after preparation. No important decrease was observed in the retention of licopene. The high retention rates showed that the evaluated procedures contributed to control the loss of carentoids in vegetables and therefore these measures should be adopted in other HFS.

En este trabajo se determinan las condiciones de temperatura más favorables para un proceso de secado de uchuva (Physalis peruviana L) con aire caliente, con deshidratación osmótica (DO) como pretratamiento, utilizando una solución de sacarosa de 70o Brix a 40oC durante 16 horas. Se realiza un seguimiento de la degradación de β-caroteno con el tiempo y la temperatura. En la fruta tratada con aire caliente a 60oC y pretratada con DO, se obtiene una pérdida total de β-caroteno del 98%. La fruta tratada con aire caliente a 40 oC y sin DO, presenta la menor pérdida total de β-caroteno, la cual alcanza un 28%. Los tiempos de secado para alcanzar una humedad de la fruta cercana a 2,5% base seca son de 7, 9 y 12 horas a 60, 50 y 40oC respectivamente, para las frutas tratadas sin DO. Para las frutas tratadas con DO, los tiempos de secado son de 4, 5 y 6 horas a 60, 50 y 40ºC respectivamente. La cinética de degradación encontrada es de primer orden. El estudio de estimación de costos de proceso de secado encuentra que la mejor condición de proceso es a 60oC y sin el tratamiento de DO, con un costo aproximado de procesamiento de $374,42/Kg de fruta.


This study find the air temperatures for dehydrating Cape gooseberry, (Physalis peruviana L), with osmotic dehydration as a pretreatment, using a solution of sucrose of 70 o Brix to 40 oC by 16 hours. The highest level of β-carotene degradation, 98%, due to the effects of oxygen, temperature and provitamin lixiviation, takes place with the pretreated fruits, which dry at 60ºC. Lixiviation contributes to 80% of this. The fruits without pretreatment and dry at a temperature of 40ºC suffer a lower level of β-carotene degradation, 28%. The times for the untreated fruit, to reach a moisture level close to 2.5%, decrease as the air temperature increase (40, 50 y 60ºC, 12, 9 and 7 hours respectively). In the case of the pretreated fruit, the drying times are 4, 5, and 6 hours for 60, 50 y 40ºC respectively. Degradation kinetic is first order. The economic study find that the cost for dehydrating at 60°C for the untreated fruit was the most suitable with an approximately cost of $374.42/Kg of fruit.

ABSTRACT

OBJETIVO: Avaliar as concentrações séricas de retinol e beta-caroteno de pré-escolares em Teresina, Piauí, com caracterização do perfil antropométrico e do consumo alimentar. MATERIAL E MÉTODOS: Estudo transversal envolvendo 135 crianças em creche municipal, com avaliação do estado nutricional pelos métodos: bioquímico (concentração sérica de retinol e beta-caroteno), antropométrico (índices de peso para estatura - P/E e estatura para idade - E/I) e dietético (freqüência de consumo alimentar). RESULTADOS: Observou-se prevalência de deficiência de vitamina A (DVA) de 8,9 por cento (IC95 por cento: 4,7 - 15,0 por cento) e existência de associação entre suplementação anterior e concentrações de retinol, com maior proporção de crianças com níveis normais de retinol entre as suplementadas (p = 0,025). As concentrações de retinol e de beta-caroteno mostraram-se correlacionadas, porém com força leve a moderada (p < 0,021). Os percentuais de crianças com baixo P/E e de baixa E/I foram de 1,9 por cento (IC95 por cento: 0,2 - 6,8 por cento) e 9,7 por cento (IC95 por cento: 4,8 - 17,1 por cento), respectivamente. Na avaliação dietética verificou-se baixo consumo de alimentos ricos em vitamina A. CONCLUSÕES: A elevada prevalência de DVA nas crianças, combinada com a alta percentagem de crianças com valores aceitáveis de retinol, os baixos valores medianos de beta-caroteno, a alta percentagem de déficit estatural e a inadequação do consumo de alimentos ricos em vitamina A, indicam a necessidade de se aprimorar as estratégias de educação em saúde e nutrição da população, incentivando o consumo de alimentos fontes de vitamina A, como medidas auto-sustentáveis importantes no combate ao problema. Além disso, deve ser considerado o incentivo à fortificação dos alimentos e ao fortalecimento de Programas de suplementação.

ABSTRACT

Since humans cannot synthesize carotenoids, they depend upon the diet exclusively for the source of these micronutrients. It has claimed that they may alleviate chronic diseases such as cancers. The present communication constitutes a global review of the scientific literature on plants and others organisms that biosynthesize carotenoids, which include the series alpha-, beta-, gamma-, delta- and epsilon-carotenes. The results of the literature survey lists more than five hundred sources.


Uma vez que os humanos não podem sintetizar carotenóides, dependem exclusivamente da dieta como fonte desses micronutrientes. Tem sido afirmado que eles podem aliviar doenças crônicas, tais como cânceres. O presente artigo constitui uma revisão global da literatura científica sobre plantas e outros organismos que biossintetizam carotenóides, que incluem as séries alfa-, beta-, gama-, delta-, e epsilon-carotenos. Os resultados das listas de pesquisa da literatura mostram mais de quinhentas fontes.

ABSTRACT

OBJETIVO: Analisar os efeitos do betacaroteno no processo de remodelação ventricular após o infarto agudo do miocárdio (IAM), em ratos expostos à fumaça do cigarro. MÉTODOS: Após o IAM, os animais foram divididos em quatro grupos: 1) grupo C, 24 animais que receberam dieta-padrão; 2) grupo BC, 26 animais que receberam betacaroteno; 3) grupo EFC, 26 animais que receberam dieta-padrão e foram expostos à fumaça de cigarro; e 4) grupo BC+EFC, 20 animais que receberam betacaroteno e foram expostos à fumaça de cigarro. Após seis meses, foi realizado estudo morfofuncional. Utilizou-se significância de 5 por cento. RESULTADOS: Em relação às áreas diastólicas (AD) e sistólicas (AS), os valores do grupo BC foram maiores que os do grupo C. Considerando a AD/peso corporal (PC) e AS/PC, os valores do grupo BC+EFC foram maiores que os valores de C. Em relação à fração de variação de área, foram observadas diferenças significativas entre EFC (valores menores) e C (valores maiores) e entre BC (valores menores) e C (valores maiores). Não foram observadas diferenças entre os grupos em relação ao tamanho do infarto. O grupo EFC apresentou valores maiores da área seccional dos miócitos (ASM) que os animais-controle. Em adição, o grupo BC+EFC apresentou maiores valores de ASM que BC, EFC e C. CONCLUSÃO: Após o infarto do miocárdio, o tabagismo e o betacaroteno promoveram intensificação do processo de remodelação cardíaca; houve potencialização dos efeitos deletérios no processo de remodelação com os dois tratamentos em conjunto.


OBJECTIVE: To analyze the effects of beta-carotene on the ventricular remodeling process following myocardial infarction (MI) in rats exposed to cigarette smoke. METHODS: After acute myocardial infarction (AMI), the animals were divided into four groups: 1) Group C, 24 animals that were given standard diet; 2) Group BC, 26 animals that were given beta-carotene; 3) Group ECS, 26 animals that were given standard diet and were exposed to cigarette smoke; and 4) Group BC+ECS, 20 animals that were given beta-carotene and were exposed to cigarette smoke. After six months, a morphofunctional study was performed. We used a 5 percent significance level. RESULTS: As regards diastolic areas (DA) and systolic areas (SA), the values for the BC group were higher than those for the C group. If DA/body weight (BW) and SA/BW are considered, the values for group BC+ECS were higher than the values for group C. As regards the fractional area change, we observed significant differences between ECS (lower values) and C (higher values) and between BC (lower values) and C (higher values). Differences between groups regarding infarction size were not observed. The ECS group presented higher values for myocyte cross-section area (MCA) than control animals. Additionally, the BC+ECS group presented higher MCA values than the BC, ECS and C groups. CONCLUSION: After myocardial infarction, smoking and beta-carotene intensified the heart remodeling process; harmful effects of the remodeling process were heightened when the two treatments were used in conjunction.
